Whole Genome Sequencing (WGS) can be used to comprehensively explore all genetic variation information in the tumor genome, helping us better understand the overall situation in the tumor genome, explore the function, clinical significance, and molecular characteristics of undeveloped genomic regions, etc; There are various types of genetic variations in the tumor genome, including common point mutations, small insertions and deletions, fusion, copy number variations, as well as large structural variations, large insertions and deletions, inversions, translocations, viral integration, etc. All of these require the use of whole genome sequencing technology combined with multi omics analysis for the molecular biology basic research of the tumor genome and the mechanism research of tumor occurrence and development.

Whole Exome Sequencing (WES) can selectively sequence the coding regions of the human genome. WES accounts for only 1% of WGS, but covers approximately 85% of pathogenic gene variations. With the reduction of sequencing costs, WES is gradually being used for clinical tumor detection. For example, by detecting genetic variations in embryonic and somatic cells, it can be used for genetic tumor risk assessment and targeted drug guidance for all cancer species, efficacy evaluation, etc; WES can also be used to detect tumor mutation burden (TMB), tumor neoantigen burden (TNB), immune recognition and killing signals, etc., based on which tumor immunotherapy plans and efficacy evaluations can be selected.

Targeted gene sequencing focuses only on the genes of interest, accurately detecting somatic and germ cell mutations in tumor samples, and is used for clinical research and diagnosis of key genes and important mutations related to tumor occurrence and development, personalized medication, etc; Targeted gene sequencing covers fewer areas and can effectively control costs, especially for low-frequency somatic mutation detection. Through deep sequencing, low-frequency mutations can be detected, such as FFPE drug guidance, liquid biopsy, etc.

By sequencing mRNA transcriptome, gene expression differences can be detected, unknown and rare transcripts can be identified, and variable splicing sites, gene fusion, SNPs, and Indel sites can be accurately identified. By sequencing non coding RNAs, such as long stranded non coding RNAs (lncRNAs) and small RNAs (Small RNAs), the types and contents of corresponding non coding RNAs, as well as their target genes, can be identified and predicted, which can be used for tumor regulation and development research.

Methylation sequencing of tumors is a method of detecting and diagnosing tumors using DNA methylation as an epigenetic marker. DNA methylation refers to the substitution of carbon hydrogen bonds on DNA molecules by carbon methyl bonds, thereby affecting gene expression and regulation. The DNA methylation pattern of tumor cells is significantly different from that of normal cells, and can be used to distinguish cancer from normal tissue, as well as determine the origin and type of cancer.
